| 摘 要：［目的］ 检测胃癌组织及相应癌旁组织中Sox7、Sox17基因的甲基化状态，探讨其与胃癌发生的关系及对预后的影响。［方法］应用甲基化特异性PCR（methylation specific PCR，MSP）、RT-PCR方法检测96例胃癌及相应癌旁组织中Sox7、Sox17基因的甲基化状态及mRNA表达情况。［结果］ 在96例胃癌组织标本中，Sox7、Sox17基因的甲基化率分别为49.0%(47/96)、78.1%(75/96)，均明显高于癌旁非肿瘤组织（P<0.01）；癌组织中Sox7、Sox17mRNA阳性表达率分别为30.2%（29/96）、14.6%（14/96），均明显低于癌旁非肿瘤组织（P<0.01）。Sox7、Sox17基因的甲基化率均与该基因的mRNA表达密切相关（P<0.01）；癌组织中Sox7、Sox17基因的甲基化状态与胃癌患者的病理分级、临床分期及淋巴结转移均无关，但两基因共同甲基化率与淋巴结转移相关。对96例胃癌患者进行随访，失访率为10.4%。Sox7基因甲基化组与非甲基化组的5年累积生存率分别为23.4%和46.9%，Sox17基因甲基化组与非甲基化组的5年累积生存率分别为30.6%和52.4%，而Sox7、Sox17基因共同甲基化组与非共同甲基化组的5年累积生存率分别为13.9%和48.3%。［结论］ 胃癌患者中Sox7、Sox17基因甲基化状态与胃癌患者的生存相关。 |

| Abstract：［Purpose］ To investigate the methylation status of Sox7 and Sox17 gene in gastric cancer (GC) and corresponding adjacent non-concerous tissue，and to analyze the relationship between Sox7，Sox17 gene methylation status and the prognosis in patients with gastric cancer.［Methods］ MSP and RT-PCR methods were applied to examine the CpG methylation of the Sox7 and Sox17 promoter and its mRNA expression in 96 samples of GC and corresponding adjacent tissues. ［Results］ In 96 samples，the Sox7 and Sox17 methylation were detected in 49.0%(47/96) and 78.1%(75/96) in tumors tissue，which were significantly higher than those in adjacent non-cancerous tissue(P<0.01). The positive expression of Sox7 and Sox17 mRNA were 30.2%（29/96），14.6%（14/96） respectively，which were significantly lower than those in adjacent non-cancerous tissue(P<0.01). Furthermore，the hypermethylation status of Sox7 and Sox17 were correlated with the expression of mRNA (P<0.01). The Sox7 and Sox17 methylation did not correlate with histologic grade ，clinical stage and lymph node metastasis. But the simultaneous methylation of Sox7 and Sox17 was associated with lymph node metastasis. Followed up 96 cases of gastric cancer，the rate of lost to follow up was 10.4%. The 5-years survival rate was 23.4% and 46.9% in Sox7 gene methylation group and non methylated group. The 5-years survival rate was 30.6% and 52.4% in Sox17 gene methylation group and non methylated group. The 5-years survival rate was 13.9% and 48.3% in the simultaneous methylation group and non simultaneous methylation group. ［Conclusion］ The methylation of Sox7 and Sox17 gene is related to survival in patients with gastric cancer. |
