      摘 要:[目的] 研究MMP10对肝癌细胞增殖的影响并探讨其分子机制。[方法] 用Western blot验证MMP10在肝癌细胞Huh-7和SMMC-7721细胞中的过表达水平;MTS分析过表达MMP10对Huh-7和SMMC-7721细胞增殖的影响;EdU标记实验检测MMP10对Huh-7和SMMC-7721细胞DNA合成能力的影响;qPCR筛选过表达MMP10对与肝癌细胞增殖有关的抑癌基因和生长因子表达的影响;MTS进一步分析该抑癌基因或生长因子在MMP10调控肝癌细胞增殖中的作用。[结果] MMP10成功过表达,在第6d过表达MMP10使Huh-7和SMMC-7721细胞的增殖能力分别增加72%和58%(P<0.001),同时过表达MMP10使Huh-7和SMMC-7721细胞的DNA合成能力分别增加81%和78%(P<0.05);MMP10促进EGFR的表达,沉默EGFR使MMP10对肝癌细胞增殖的促进作用降低81%(P<0.01)。[结论] MMP10促进肝癌细胞增殖,下调EGFR的表达拮抗MMP10对肝癌细胞增殖的促进作用。

      Abstract:[Purpose] To investigate the effect of matrix metalloproteinase 10(MMP10) on hepatocellular carcinoma(HCC) cells proliferation and its molecular mechanism of MMP10 regulation on HCC cells proliferation. [Methods] The overexpression level of MMP10 in Huh-7 and SMMC-7721 cells was verified by Western blot,the effect of overexpression MMP10 to Huh-7 and SMMC-7721 cells proliferation was analyzed by MTS,the effect of overexpression MMP10 to DNA synthesis of Huh-7 and SMMC-7721 cells was examined by EdU incorporation assay,the expression of tumor suppressor genes and growth factors associated with HCC cells proliferation was detected by qPCR after overexpression MMP10,the role of tumor suppressor gene and growth factor in MMP10 regulation on the HCC cells proliferation was analyzed by MTS. [Results] The overexpression of MMP10 was successful. The proliferation of Huh-7 and SMMC-7721 cells after overexpression MMP10 for 6 days increased 72% and 58%,respectively(P<0.001). Moreover,the DNA synthesis of Huh-7 and SMMC-7721 cells increased 81% and 78%,respectively(P<0.05). In addition,MMP10 significantly promoted EGFR expression. The effect of MMP10 to HCC cells proliferation reduced 81% induced by EGFR knockdown(P<0.01). [Conclusion] MMP10 promoted the proliferation of HCC cells and EGFR attenuated the promotion of MMP10 in HCC cells proliferation.
