      摘 要:[目的] 探讨siRNA沉默酵母基因Noblp人类同源基因(NOB1)对胃癌细胞凋亡及顺铂化疗敏感性的影响。[方法] 胃癌细胞MGC-803转染NOB1 siRNA和siRNA control为干扰组、对照组,同时以顺铂处理的两组细胞为顺铂+干扰组、顺铂组,qRT-PCR和Western blot法检测NOB1表达,噻唑蓝法(MTT)检测细胞增殖,流式细胞仪检测细胞凋亡,Western blot检测信号转导与转录因子3(STAT3)、磷酸化STAT3(p-STAT3),比色法检测含半胱氨酸的天冬氨酸蛋白水解酶3(Caspase-3)活性。[结果] 干扰组、顺铂组、顺铂+干扰组细胞中NOB1 mRNA和蛋白水平下降,细胞存活率降低,细胞凋亡率升高,细胞中Caspase-3活性升高,p-STAT3/STAT3水平降低,与对照组相比差异具有统计学意义(P<0.05)。顺铂+干扰组细胞凋亡率和Caspase-3活性明显高于干扰组、顺铂组,细胞存活率和p-STAT3/STAT3水平明显低于干扰组、顺铂组,差异均具有统计学意义(P<0.05)。[结论] siRNA沉默NOB1基因促进胃癌细胞凋亡,增加顺铂敏感性,作用机制与p-STAT3/STAT3有关。

      Abstract:[Purpose]To investigate the effect of siRNA silencing NOB1 gene on apoptosis and chemosensitivity to cisplatin in gastric cancer cells. [Methods] Gastric cancer MGC-803 cells were transfected with NOB1 siRNA or siRNA control,then treated with or without cisplatin;thus the cells were divided into 4 groups:interference,control,interference+cisplatin and cisplatin groups. The expression of NOB1 was detected by RT-PCR and Western blot;cell proliferation was detected by MTT,apoptosis was detected by flow cytometry,the expression of STAT3 and p-STAT3 was detected by Western blot,and Caspase-3 activity was detected by colorimetric assay. [Results] The levels of NOB1 mRNA and protein decreased,cell viability decreased,the rate of apoptosis was increased,Caspase-3 activity was elevated,levels of p-STAT3/STAT3 were decreased in interference,interference + cisplatin and cisplatin groups,compared with the control group(all P<0.05). The apoptosis rate and Caspase-3 activity of interference + cisplatin group were increased,and the cell viability and p-STAT3/STAT3 levels were decreased more markedly than those in interference group and cisplatin group(P<0.05). [Conclusion] Silencing NOB1 gene by siRNA promotes apoptosis of gastric cancer cells and increases their sensitivity to cisplatin,which may be related to p-STAT3/STAT3 pathway.
