      摘 要:[目的] 分析Kiaa0247与肺癌患者临床特征及肺癌细胞增殖和凋亡的相关性。[方法] 免疫组织化学染色法检测2018年1月至2019年12月收治的肺癌患者病理组织中Kiaa0247的表达,并分析其表达与临床因素的相关性。实时荧光定量PCR检测正常肺上皮细胞BEAS-2B及不同肺癌细胞系中Kiaa0247的表达水平,选取表达量最高的细胞作为研究对象,再将Kiaa0247 si-RNA和 Kiaa0247 质粒转染至A549细胞,沉默或过表达细胞中的Kiaa0247,MTT法检测细胞增殖,流式细胞仪测细胞周期及细胞凋亡情况,Western blot方法检测Bax及Bcl-2的蛋白表达。[结果] Kiaa0247在肺腺癌组织中表达低于正常肺组织(1.14 vs 0.69,P<0.05),鳞癌中的表达水平无明显差异(1.14 vs 1.22,P>0.05)。Kiaa0247表达量与患者TNM分期及有无淋巴结转移明显相关。与BEAS-2B细胞相比,A549细胞系中Kiaa0247表达量最高(1.52 vs 0.84,P<0.01),Kiaa0247 si-RNA可降低Kiaa0247的蛋白(0.54 vs 0.23,P<0.01)及RNA表达(0.85 vs 0.43,P<0.01),转染质粒可上调Kiaa0247的蛋白(0.63 vs 0.96,P<0.01)及RNA表达(0.76 vs 1.31,P<0.01)。沉默Kiaa0247在48h及72h时促进A549的增殖(0.53 vs 0.73,P<0.05)(1.21 vs 1.57,P<0.01),过表达Kiaa0247可明显抑制A549细胞增殖(0.86 vs 0.70,P<0.05) (1.51 vs 0.85,P<0.01),主要通过细胞G1/S 期的转变影响增殖。沉默Kiaa0247后抑制细胞凋亡,且使Bax表达量明显降低(1.23 vs 0.78,P<0.01),Bcl-2表达明显升高(0.56 vs 1.35,P<0.01),上调Kiaa0247促进细胞凋亡,Bax表达明显升高(0.68 vs 1.32,P<0.01),Bcl-2明显降低(1.12 vs 0.54,P<0.01)。[结论] Kiaa0247表达可反应肺癌患者的临床分期及淋巴结转移情况,其表达量的高低明显影响肺癌细胞增殖及凋亡。

      Abstract:[Purpose] To investigate the expression of Kiaa0247 in lung cancer cells and its relation with clinical feature,cancer cell proliferation and apoptosis. [Methods] Immunohistochemical staining was used to detect the expression of Kiaa0247 in cancer and normal lung tissue samples from 120 patients with lung cancer,including 49 cases of lung adenocarcinoma and 71 cases of lung squamous cell carcinoma,treated in our hospital from January 2018 to December 2019,the correlation between its expression level and clinicopathological factors was analyzed. The level of Kiaa0247 in human lung epithelial cells BEAS-2B and lung cancer cell lines were detected by qRT-PCR. The cells with the highest expression level of Kiaa0247 were selected as the research objects. Kiaa0247 si-RNA and Kiaa0247 plasmid were transfected into lung cancer A549 cells to silence or over-express Kiaa0247. Then cell proliferation was detected by MTT assay,cell cycle and apoptosis was examined by flow cytometry. The expression of Bax and Bcl-2 were measured by Western-blot. [Results] The expression of Kiaa0247 in lung adenocarcinoma tissues was lower than that in normal lung tissues(1.14 vs 0.69,P<0.05),and there was no significant difference in the expression level in squamous cell carcinoma(1.14 vs 1.22,P>0.05). Kiaa0247 expression was significantly related with TNM staging and lymph node metastasis. Compared with BEAS-2B cells,the expression level of Kiaa0247 was the highest in A549 cell line(1.52 vs 0.84,P<0.01). Kiaa0247 Si-RNA decreased Kiaa0247 protein(0.54 vs 0.23,P<0.01) and RNA expression(0.85 vs 0.43,P<0.01),while transfection of Kiaa0247 plasmid increased Kiaa0247 protein(0.63 vs 0.96,P<0.01) and RNA expression(0.76 vs 1.31,P<0.01). Silencing Kiaa0247 promoted the proliferation of A549 by promoting G1/S phase transition at 48h(0.53 vs 0.73,P<0.05) and 72h(1.21 vs 1.57,P<0.01). Overexpression of Kiaa0247 significantly inhibited the proliferation of A549 cells at 48h(0.86 vs 0.70,P<0.05) and 72h(1.51 vs 0.85,P<0.01). Silencing of Kiaa0247 was able to inhibit apoptosis of A549 cells and change the expression of Bax(1.23 vs 0.78,P<0.01) and Bcl-2(0.56 vs 1.35,P<0.01).[Conclusion] Kiaa0247 expression can reflect the clinical stage and lymph node metastasis of lung cancer patients,and its expression levels significantly affect the proliferation and apoptosis of lung cancer cells.
