| [目的] 探讨NLRP3在胃癌中的表达及与预后的相关性。[方法] 使用GEO数据库行COX回归分析评估NLRP3及临床病理因素与生存率的相关性，采用免疫组化检测胃癌及癌旁组织中NLRP3蛋白表达，Kaplan-Meier曲线分析不同NLRP3表达水平总生存期的差异。[结果] GSE183136数据集567例胃癌单因素COX回归分析显示，临床分期、年龄、脉管侵犯及NLRP3与胃癌预后相关（P<0.05），多因素COX回归分析表明，低表达NLRP3是生存的独立危险因素（HR=1.39[1.04，1.89]，P=0.027）。112例癌组织NLPR3免疫组化评分均值（3.49±3.16）低于癌旁组织（5.54±3.51，P<0.001），NLRP3表达阴性与年龄、T分期及TNM分期相关（P<0.05），预后差于阳性表达者（HR=2.34[1.04-5.30]，P=0.041）。[结论] 低表达NLRP3是胃癌生存的危险因素，免疫组化表达阴性可能作为胃癌不良预后的预测指标。 |

| [Objective] To investigate the expression of NLRP3 and the correlated with prognosis in gastric cancer. [Methods] COX regression analysis was performed to evaluate the correlation among NLRP3, clinicopathological factors and overall survival (OS) with dataset from the GEO database. The expression of NLRP3 protein in cancer and para-cancerous samples was measured by immunohistochemistry. The Kaplan-Meier curve was draw to show the OS with different NLRP3 expression levels. [Results] Univariate COX regression analysis of 567 patients from GSE183136 dataset showed that clinical stage, age, lympho-vascular invasion and NLRP3 were associated with prognosis of gastric cancer (P<0.05). The multivariate COX regression analysis investigated that low expression of NLRP3 was an independent risk factor for death (HR=1.39 [1.04, 1.89], P=0.027). The mean NLPR3 immunohistochemical intensity score of 112 cases of cancerous tissues (3.49±3.16) was lower than that of para-cancerous tissues (5.54±3.51, P<0.001). Negative expression of NLRP3 in IHC stain was associated with age, T stage and TNM stage (P<0.05), and the prognosis was worse than that of positive expression (HR=2.34[1.04-5.30]，P=0.041). [Conclusion] Low expression of NLRP3 is a risk factor for gastric cancer survival, and negative expression of immunohistochemical stain may be used as a poor prognostic marker for gastric cancer. |
